Por ello, la identificaci\u00f3n de tratamientos, pero sobre todo protocolos para su predicci\u00f3n y prevenci\u00f3n, se han convertido en prioridades para la investigaci\u00f3n en biomedicina y para su traducci\u00f3n a la pr\u00e1ctica cl\u00ednica. Estas enfermedades tienen una etiolog\u00eda compleja, con una importante carga gen\u00e9tica y una fuerte dependencia de variables externas, ambientales, de estilo de vida y del exposoma. Numerosos estudios gen\u00e9ticos han identificado cientos de variaciones en el genoma asociadas al riesgo de desarrollar este tipo de enfermedades, con la promesa y esperanza de que sus resultados permitan identificar dianas farmacol\u00f3gicas y la posibilidad de predecir el riesgo de padecer la enfermedad. Sin embargo, aunque se han dirigido a genes espec\u00edficos, la traducci\u00f3n de esta informaci\u00f3n gen\u00e9tica en modelos predictivos con validez cl\u00ednica es actualmente muy limitada. Esto se debe al escaso poder predictivo de los enfoques reduccionistas, como los Polygenic Risk Scores, y a las limitadas posibilidades de integrar otras variables cl\u00ednicas, ambientales y de estilo de vida, que son potenciales modificadores del riesgo en este tipo de patolog\u00eda. Por ello, en este proyecto proponemos la construcci\u00f3n de modelos de predicci\u00f3n de la diabetes tipo 2, basados en enfoques m\u00e1s integradores utilizando inteligencia artificial (modelos de aprendizaje autom\u00e1tico), para la identificaci\u00f3n de determinantes gen\u00e9ticos, cl\u00ednicos (variables y patolog\u00edas asociadas) y ambientales, y para la construcci\u00f3n de modelos de predicci\u00f3n y clasificaci\u00f3n de pacientes con diabetes tipo 2. El proyecto IA4DT2 tiene como\nobjetivo desarrollar modelos predictivos de DT2 basados en datos gen\u00e9ticos, ambientales,\nde estilo de vida y cl\u00ednicos, adem\u00e1s de identificar marcadores epigen\u00e9ticos clave en su\ndesarrollo.\n\nEn colaboraci\u00f3n entre el grupo de Gen\u00e9tica Computacional del Barcelona Supercomputing\nCenter, el grupo GCAT|Genomes for Life del Institut Germans Trias i Pujol y el departamento\nde Derecho P\u00fablico de la Universidad del Pa\u00eds Vasco, se han implementado protocolos para\nfacilitar el acceso y transferencia de datos. Esto permiti\u00f3 el an\u00e1lisis de las cohortes de UK\nBiobank y de GCAT, con informaci\u00f3n gen\u00e9tica, de estilo de vida y cl\u00ednica de m\u00e1s de 500,000\nparticipantes. Se generaron y validaron distintos modelos predictivos de DT2 (gen\u00e9tico,\ncl\u00ednico y de estilo de vida) con un poder de detecci\u00f3n entre 60-73%, mejorando la detecci\u00f3n\nde la predisposici\u00f3n gen\u00e9tica a DT2 y proponiendo nuevos objetivos de estudio en\nmarcadores gen\u00e9ticos y ambientales.\n\nAdicionalmente, el an\u00e1lisis de patrones epigen\u00e9ticos en GCAT permiti\u00f3 la identificaci\u00f3n de\nregiones y genes clave en la regulaci\u00f3n de la glucosa y resistencia a la insulina.
