En a\u00f1os recientes, los f\u00e1rmacos degradadores han demostrado que la proximidad inducida a la maquinaria celular de destrucci\u00f3n proteica (E3 ubiquitina ligasas) puede provocar la ubiquitinaci\u00f3n y degradaci\u00f3n de prote\u00ednas diana, convirti\u00e9ndose as\u00ed en una alternativa prometedora para la intervenci\u00f3n terap\u00e9utica. Los degradadores se basan en la mera uni\u00f3n a prote\u00ednas en lugar de en su inhibici\u00f3n y, por lo tanto, prometen ampliar el proteoma atacable farmacol\u00f3gicamente. Descubrimientos fortuitos han demostrado el inmenso potencial de los degradadores conocidos como &#8220;molecular glues&#8221; (MGs). Estos MGs son compuestos que producen interacciones directas entre una E3 ubiquitina ligasa y una prote\u00edna diana, siendo capaces de eliminar dianas terap\u00e9uticas que de otro modo ser\u00edan imposibles de atacar farmacol\u00f3gicamente. Ya existen MGs en la cl\u00ednica (lenalidomida y sus an\u00e1logos), sin embargo, el descubrimiento de los MGs ha sido en gran parte fortuito. \u00bfPodemos pasar de la serendipia al dise\u00f1o racional? En los \u00faltimos a\u00f1os, encontramos MGs con un modo de acci\u00f3n sin precedentes contra la prote\u00edna ciclina K, previamente considerada imposible de atacar farmacol\u00f3gicamente. En este proyecto, esperamos continuar explorando la impresionante farmacolog\u00eda de este tipo de f\u00e1rmacos y as\u00ed ayudar a ampliar el espacio atacable del proteoma humano. In recent years, molecular glue degraders (MGs) have\ndemonstrated that compound-induced proximity to E3 ubiquitin ligases can prompt the\nubiquitination and degradation of disease-relevant proteins, providing a promising alternative for\ntherapeutic intervention. Rather than relying on inhibition, these degraders function by binding\nproteins and orchestrating their elimination, thereby broadening the druggable proteome. While\nclinical proof for this approach already exists, as exemplified by lenalidomide and its analogs\n(currently used against multiple myeloma), the discovery of MGs has historically been largely\nserendipitous, limiting their broader application as a generalizable drug solution.\nThis project has addressed this challenge by developing a systematic platform to move MG\ndiscovery from serendipity to intentional design. By employing innovative screening strategies that\ninterrogate E3 ligase activity, we have identified some chemical matter that can be classified as\nMG-like compounds. Our approach has yielded promising results, demonstrating the feasibility of\nuncovering new classes of degraders with anticancer potential and expanding the range of proteins\nthat can be targeted therapeutically.
